Denatured, not killed: the enzyme word the mark scheme refuses

It is the most repeated piece of advice in GCSE biology, and it is usually given without the evidence. Here is the line in the mark scheme, twice, and why the board draws it where it does.

The line in the mark scheme

Teachers say it every year and students half believe it: write denatured, never killed. The advice is right, and it deserves better than to be taken on trust, because the board wrote it down.

AQA’s mark scheme for GCSE Biology Paper 1, Higher Tier, from the November 2020 series, asks in question 01.8 what happens to an enzyme at high temperature. The credited answer is that the enzyme loses the shape of its active site. Beside it, in the column examiners read, are three allowances and one refusal.1

A year later the same paper asked about amylase at 80 °C, and the scheme says the same thing in slightly different words: the credited answer is that “the amylase has been denatured”, and the refusal is “do not accept the amylase is killed / has died”.2

Two series, two questions, one line. It is a real rule, it is written down, and a reader can open the PDF and find it in under a minute.

Why an enzyme cannot be killed

Look at what the November 2020 scheme allows before it refuses. “Destroyed” is allowed. “Damaged” is allowed. Neither is the technical word, and neither says anything about a shape. So the line is not “use the specialist term or score nothing”. The line is narrower and more interesting than that: do not say something false.

An enzyme is a protein. It is a molecule, and a molecule was never alive. “Killed” is a claim about an organism: something that was living and now is not. Applied to an enzyme it is not vague, the way “damaged” is vague. It is wrong, in the way that “the water died” is wrong. And a student who writes it has told the marker something about what they think an enzyme is, which is the thing the question was quietly testing.

That is why “destroyed” survives and “killed” does not. Destroying a molecule is a thing that can happen. Killing one is not.

What denatured actually says

The word earns its place because it carries the mechanism in one move. A protein’s shape is held by bonds between parts of its own chain. Heat, or a pH far from the one it works at, breaks those bonds, and the chain settles into a different shape. For an enzyme, the shape that mattered was the active site, and once that has changed the substrate no longer fits it: it is no longer complementary, no enzyme-substrate complex forms, and the reaction stops.

Every clause of that paragraph is a marking point somewhere on a GCSE paper, and “denatured” is the one word that stands for all of them. That is also why the full-mark answer at GCSE is the longer form, “loses the shape of the active site”: it says what denaturing does, rather than only that it happened. If there are two marks on offer, give the word and the shape.

One more thing the word says, which students rarely use: a denatured enzyme is permanently changed. Cooling it down does not fold it back. An enzyme that has simply slowed down in the cold has not been denatured at all, and writing “denatured” for a cold enzyme is a different mistake with the same cause.

The same rule a level up

At A-level the word is assumed and the scrutiny moves to what surrounds it. AQA’s June 2022 Paper 1 report, on a question where students had to read a graph of enzyme activity, records that “many students wrote that an enzyme ‘worked well’, which was insufficient at this level for an indication of enzyme activity”.3 The thing that changed has a name, rate of reaction or enzyme activity, and a verdict on it (“well”, “badly”) is not a measurement of anything.

The same report shows the word doing a second job that almost nobody gave it. Asked how to prepare a control for an enzyme investigation, students overwhelmingly changed the independent variable instead, and “references to using a denatured enzyme were rarely seen”.3 A boiled enzyme is the control: the same protein, the same volume, the same everything, except that it cannot work. The word that GCSE spends a mark on is, at A-level, the answer to a practical question worth three, and students who had it did not think to use it.

Credited and refused

From the two mark schemes and the one report cited, all AQA. The allowances in the first row are exactly what the November 2020 scheme printed; another series may allow differently, and the safe answer is the credited one.
